The hydrolysis of phosphate esters of alpha-hydroxy acids catalyzed by molybdate.

چکیده
Frequent observations have been made of the increased rate of hydrolysis of relatively labile phosphate compounds in acid molybdate solution (e.g. phosphocreatine (I), acetyl phosphate (2), and 1,3-diphosphoglyceric acid (3)). A study by Weil-Malherbc and Green (4) indicated that ATP, pyrophosphate, phosphoenolpyruvate, and fructosr B-phosphate were hydrolyzed appreciably in the pH range 3 to 5, while fructose 1,6-diphosphate, glucose lphosphate, cY-plycerophosphatc, and glucose B-phosphate were quite stable. The fact that monophosphoglyceric acids form complexes with high optical rotation in the presence of molybdate led to the use of polarimetric procedures for the quantitative determination of these compounds (5). Alolybdate forms complexes with man3 oxygen-containing organic compounds and the affinity for a-hydroxy acids is particularly high (6). The present study began with the observation of Bartlett that the monophosphoglyceric acids are hydrolyzed at widely different< rates in the presence of molybdate (7). The specificity, pH, and temperature dependences have been examined. In the pH range 3 to 5 there is a marked specificity of hydrolysis for phosphate esters of oc-hydroxy acids. A procedure has been worked out for the quantitative

متن کامل1-Deoxy-D-xylulose: synthesis based on molybdate-catalyzed rearrangement of a branched-chain aldotetrose.
1-Deoxy-D-xylulose has been prepared in seven steps and approximately 21% overall yield from 2,3-O-isopropylidene-D-erythrono-1,4-lactone. The key reaction involves transformation of a branched-chain aldotetrose to the 1-deoxy-2-ketopentose catalyzed by molybdic acid. Other branched-chain aldotetroses containing bulkier substituents at C2 also engage in the conversion, suggesting routes to prot...
متن کاملThe catalytic effect of molybdate on the hydrolysis of organic phosphate bonds.
The great instability of creatine phosphate (Fiske & Subbarow, 1929), acetyl phosphate (Lipmann & Tuttle, 1944) and 1:3-diphosphoglyceric acid (Negelein & Bromel, 1939) in solutions containing molybdate has been known almost as long as these compounds themselves. It could not fail to attract the attention of the analyst who used a method of phosphate estimation ofthe Briggs (1922) or Fiske & Su...
متن کاملThe catalytic effect of molybdate on the hydrolysis of organic phosphates.
The increased rate of hydrolysis of organic phosphate compounds in the presence of molybdate has been observed frequently and is the basis for the estimation of phosphocreatine (1) and acetyl phosphate (2) in tissue extracts. A study was undertaken to determine whether this was a general effect and, if so, to elucidate the mechanism. While these experiments were in progress, Weil-Malherbe and G...
متن کاملA peroxidase-coupled method for the colorimetric determination of serum triglycerides.
We describe an enzymatic method for rapid, precise measurement of serum triglycerides with use of sample:reagent ratios as large as 1:200. Hydrolysis of triglycerides is catalyzed by lipase to produce glycerol and free fatty acids. The glycerol generated is then phosphorylated by adenosine 5'-triphosphate in the presence of glycerol kinase. Oxidation of the resulting glycerol 3-phosphate to pro...
